Retatrutide Phase 3 Trial: Adults With Type 2 Diabetes Lost Average of 50 Pounds

Eli Lilly and Company has reported detailed Phase 3 trial results for retatrutide, an investigational medication that produced significant weight loss and improvements in blood sugar control among adults with obesity or overweight and type 2 diabetes. Results from the study were presented at the annual meeting of the European Association for the Study of Diabetes and simultaneously published in The Lancet.

The findings come from the TRIUMPH-2 trial, which enrolled 1,152 adults with obesity or overweight and type 2 diabetes. Participants were randomly assigned to receive weekly injections of retatrutide at doses of 4 mg, 9 mg or 12 mg, or placebo, for 80 weeks.

According to Lilly, participants receiving retatrutide experienced average weight reductions of 12.7%, 19.1% and 20.8% across the three dose groups, compared with 4.0% for placebo. Participants receiving the highest dose lost an average of 49.6 pounds over the 80-week study period.

Among participants with a baseline body mass index of 35 or greater, those receiving retatrutide 12 mg lost an average of 23.4% of their body weight. Lilly also reported that 59.5% of participants receiving the highest dose no longer met BMI criteria for obesity at the end of the trial.

The study also evaluated glycemic control. Participants receiving retatrutide reduced A1C by an average of up to 1.6%, while up to 40% achieved an A1C below 5.7%.

Retatrutide is an investigational, first-in-class GIP, GLP-1 and glucagon triple hormone receptor agonist. The therapy has not been approved by the FDA and is not currently available for prescribing outside clinical trials.
